I have two projects:
1) Quantitative comparison between exosomes and microvesicles of wild-type KRAS expressing colon cancer cells utilizing iTRAQ. In this project, I have found an enrichment of proteins associated with cell-cell junctions, cell-matrix adhesion, exosome biogenesis and cell signaling in small EVs/exosomes.
2) KRAS-regulated Ago2-mediated sorting of extracellular RNAs into exosomes- I am using the expression of Ago2 phosphomutants in mutant KRAS-expressing colon cancer cells as a tractable system to identify Ago2-driven RNA secretion in exosomes. Additionally, in this project, I will determine the functional consequence of Argonaute-mediated RNA secretion for KRAS-driven colon tumors.
