Notch signaling is required for the formation of mesangial cells from a stromal mesenchyme precursor during kidney development.

Nephrogenesis, or the development of functional units within the kidney, requires two distinct populations of mesenchyme: cap mesenchyme (CM) that gives rise to epithelial cells, and stromal mesenchyme (SM) that gives rise to endothelial cells.  The molecular processes required for CM development are relatively well understood, whereas those for SM are not.  Notch signaling is necessary for normal kidney development (mouse hylomorphic Notch2 models), and here fate mapping and knock-out mouse models indicate a specific role for Notch in the development of vascular smooth muscle components such as mesangial cells from surrounding SM.
