G²³ activated by various upstream Gi/o-coupled G protein coupled receptors binds to t-SNARE protein complexes that anchor vesicles to the membrane of presynaptic neurons. This interaction inhibits exocytosis in neurons. While residues important for binding on both Gβγ and the t-SNARE complex have been identified, the binding site remains uncharacterized. Crosslinking and x-ray crystallographic studies will be conducted to provide structural and biochemical insight into this physiologically relevant interaction.
