Internship: Intern – Research and Early Development – Structural Biology, Genentech, San Francisco, CA

Start Date: Summer 2019

Length of Assignment: 12 weeks

Work Hours: 40 hours per week

The Structural Biology mission is to collaboratively advance drug discovery through understanding of the structure, function and regulation of proteins of therapeutic interest.

The Structural Biology department is looking to hire three interns for the following projects.

Project 1

The intern will be highly motivated scientifically and work in the group to do following:

  • Express proteins in E coli
  • Purify Proteins using affinity and other chromatography methods
  • Collaborate to biophysically / structurally characterize proteins

The goal of the project is to introduce symmetry in monomeric proteins by designing mutations, and testing to verify the introduced symmetry by biophysical characterization methods such as Crystallography or Electron Microscopy. This would be a collaborative project and the intern will become familiar with cutting edge facilities in-house for structural biology and protein expression.

Qualifications
  • Team Player with Fearless Attitude
  • BS or MS in Chemistry or Biology
  • Familiarity with protein biochemistry such as protein expression in E coli, protein purification and characterization is essential

Date: 01/28/19

Some experience in protein crystallization or other biophysical/biochemical characterization of proteins is preferred

Project 2

The successful intern candidate works well in small teams and is interested in pursuing basic research in drug discovery environment studying the mechanisms of assembly and regulation of challenging macromolecular complexes using biochemical and cryo-electron microscopy (cryo-EM) techniques.

The intern will engage directly in:

  • Sample purification, preparation and optimization of recombinantly produced multiprotein/ DNA complexes
  • Biochemical and structural analyses of multi-protein/DNA complexes
  • Structure determination by single particle cryo-EM (grid preparation, data analysis, etc.)
Qualifications
  • MS or pursuing PhD in Structural Biology, Biochemistry, Biophysics, or related areas
  • Experience purifying recombinant proteins, and the reconstitution and biochemical analysis of multi-protein/DNA complexes is desirable
  • Demonstrated interest and knowledge of biochemical and structural analyses of macromolecular assemblies

Project 3

The intern will explore the structure and regulatory properties of proteins of therapeutic interest. The successful candidate will be primarily responsible for the purification and characterization of large macromolecular protein complexes for structural analysis by X-ray crystallography and/or cryoEM. The work includes optimizing protein purification strategies, solution properties, and crystallization.

Qualifications
  • BS, MS, or PhD students currently studying Biochemistry, Biophysics or a related discipline
  • Strong understanding of protein biochemistry and some previous experience with recombinant protein purification using an AKTA FPLC.
  • General understanding of various structural biology techniques will be considered an asset.
  • Self-motivated, capable of working independently, and enjoy working in a collaborative setting
  • Strong analytical, communication, and organizational skills are also required
